Chadwick Boseman (T'Challa, also known as Black Panther) underwent an intense training program to get into the character's fighting style. Although he was not much of a comic book reader in his childhood, Boseman read as many Black Panther comics as possible. Boseman also reused the Xhosa accent he learned from the film Message from the King (2016) for the character.

Costume designer Judianna Makovsky explained that for all of Chris Evans's scenes in normal clothes (outside the suit), they deliberately dressed him in T-shirts several sizes smaller than his actual size so that his muscles would show on camera. Evans said all the T-shirt scenes were scheduled at the start of filming because he had lost a great deal of weight over the months leading up to shooting a Marvel film. He said that filming the action scenes in particular while wearing the Captain America costume caused him to lose a large part of his bulk.

Marvel initially wanted to hire Robert Downey Jr. to reprise Tony Stark in a small role with only three weeks of work. But Downey wanted a bigger role that would lead to a bigger payday. Marvel Entertainment CEO Ike Perlmutter was so angered by this demand that he ordered the screenwriters to remove Tony Stark from the script entirely. When the deal appeared to be off the table, Marvel Studios President Kevin Feige pushed to hire Downey, suggesting that the film's cast could open the door to sequels, new series and dramatic possibilities in the Marvel Cinematic Universe, because this film could drive the future stories of those films. As a result of Downey's inclusion in the cast, the actor received a substantial payment, including a back-end participation deal and another payment if the film's box office gross exceeded that of Captain America: The Winter Soldier (2014), which it did.
