The series underwent major changes between its first and third seasons. Although it always drew fairly high ratings, test audiences responded more strongly to the supporting characters than to the main cast. Most of the main cast was dropped in the second season (with the exception of Arye Gross), and the character Audrey Penney, who had previously appeared in only one episode, became a regular along with Joe Ferrell (David Anthony Higgins).

Unlike most TV shows, which reuse a single establishing shot throughout their run, Ellen never reused its "establishing shot" footage. Whether it was the exterior of Ellen's house or the entrance to Buy the Book, the show always aired newly filmed footage, creating a sense of variety by shooting on different days, at different times (night or day), and from different angles, with a wide lens or in close-up, with or without a zoom, and with different passersby.

After the premiere of Buffy the Vampire Slayer (1997) season 4, episode 19, "New Moon Rising," in which the ordinary character Willow comes out as a lesbian and chooses to stay with her girlfriend Tara, a group of internet message board posters sent an embossed toaster to "Buffy" creator Joss Whedon to thank him for the story. This was a nod to season 4, episode 22, "The Puppy Episode," the "coming out" episode of this series (which longtime Buffy writer Jane Espenson also worked on), which included a joke about rewarding everyone newly "joined" to the LGBT movement with a toaster.

Many episodes opened with Ellen DeGeneres introducing an artist such as ZZ Top or Sharleen Spiteri of the band Texas, who performed the theme song that week while holding up a sign with the series title on it.

Although Maggie Wheeler appeared in the main credits of every episode of season 1, she appeared in fewer than half of them.

The Canadian brand Naya was the official water of "Ellen" and can be seen in the hands of Ellen and other characters throughout the series. It is in fact a company owned and operated by Québec. From season 2 onward, they mostly drank from neat cobalt blue glass bottles, but this time the brand of water was not shown.

Ellen's real-life brother and mother, Vance and Betty DeGeneres, appeared as guest stars on the series, as did her former girlfriend Anne Heche.

In season 4, before the notorious "coming out" episode, many episodes contained hints that Ellen was gay, such as Ellen emerging from an actual closet when she buys a house and Joe asking her whether she feels she has to hide a relationship.
