In the comics, Jessica Jones attended high school with Peter Parker (a.k.a. Spider-Man), and her father worked for Tony Stark (a.k.a. Iron Man). Jones and Luke Cage are married and have a daughter named Danielle, inspired by Danny Rand/Iron Fist, a character from a different Marvel/Netflix series. Jones is also shown to have a physical attraction to Johnny Storm, AKA The Human Torch.

Malcolm Ducasse, played by Eka Darville, is Jessica Jones's neighbor struggling with drug addiction, and as a result his personal journey becomes intertwined with hers. Jessica meets him while rescuing him from two muggers; she does not remember the encounter because it was also the first night Kilgrave took control of her. In the comics, the character is a high school student named Malcolm Powder who is a Jessica Jones fan and asks for a job at Alias Investigations.

The series' world was initially going to be closer to the comics world, where superheroes were already a well-known and established part of the universe and superpowers were much more common. This was changed to match the more subdued Marvel Cinematic Universe, where the only superheroes publicly known at the time were mostly the Avengers and Daredevil. The public is nonetheless aware that there are at least more "gifted" individuals than those mentioned above.

Patsy "Trish" Walker, played by Rachael Taylor, is Jessica Jones's adoptive sister and best friend; she is a former child star of the in-universe series "It's Patsy" and the host of the radio program "Trish Talk." In the comics, Patsy Walker is one of Marvel's oldest characters. She was originally a teenage model, later reinvented as the superhero Hellcat, but had no connection to Jessica Jones.

The villain Kilgrave, played by David Tennant, was known as the Purple Man in the Marvel comics. In the comics he is Zebediah Killgrave, a spy born in Croatia who was in an accident involving chemicals that gave him permanent purple skin and powers. In this series he is portrayed as an Englishman named Kevin Thompson who gained his powers through experiments in his youth and gave himself the name Kilgrave. He has normal-colored skin but wears purple throughout the series because it is his favorite color. The scene lighting is also purple whenever Jessica is under his spell or having nightmares or flashbacks about him. Kilgrave gives Jessica a purple dress in A. K. A. WWJD? (2015).

In May 2015, Marvel announced plans to reprint "Alias," the comics series on which the show is based, with new covers by the comic's original cover artist David Mack, who also provided the illustrations for the series' opening title sequence. The reprints, released entirely digitally in June 2015 and running up to issue 15 in two trade paperbacks in September, were intended both to celebrate Jessica Jones's history and to introduce the character to new viewers ahead of the season's premiere.
