Pose stands out as the production with the most trans actors in a scripted series in history. Trans writers and producers shaped the stories by drawing inspiration from their own lives. Most of the characters are inspired by real-life trans women and ballroom mentors. Pray Tell is largely inspired by the iconic host and ballroom historian Junior Lebeija, while Blanca Evangelista is based on legendary ballroom mothers Angie Xtravaganza and Avis Pendavis. Angel's story is connected to Octavia St Laurent, who competed in and won a modeling contest organized by Eileen Ford; it also references the life of trans model Tracey Africa Norman, who was exposed and ostracized in the fashion industry. Carmen Xtravaganza was a trans model who achieved a success similar to Angel's modeling in Europe at the end of season 2. Willi Ninja inspires Damon as a vogue dancer, house father, choreographer and model. Elektra Abundance is inspired by Crystal Lebeija, down to her tone of voice. Dorian Corey may also have contributed to Elektra's story as a drag queen; a mummified body with a bullet in its head was found in her apartment, believed to be a client or boyfriend who resorted to violence. Candy Ferocity is most likely inspired by Venus Xtravaganza, a trans sex worker killed in a New York hotel.

In season 1, Pray Tell judges Angel's look in the "High Fashion Evening Wear; Luxury Ladies" category. When Angel says her dress is Halston, Pray Tell replies, "Halston?.. For JCPenney?" Halston was a popular American designer who rose to fame in the 1970s. His billion-dollar licensing deal with JCPenney in the 1980s symbolizes the brand's decline and loss of its privileged position. Pose creator Ryan Murphy made a miniseries about Halston's life for Netflix in 2021.

Pose's transgender writers and producers use their own lives as inspiration for the storylines. Many of the series' characters are inspired by real-life trans women and ballroom mentors. Pray Tell is largely inspired by the iconic host and ballroom historian Junior Lebeija. Blanca Evangelista is inspired by Angie Xtravaganza and Avis Pendavis, both legendary ballroom mothers. Angel has a connection to Octavia St Laurent, who, like Angel, signed up for a modeling competition run by Eileen Ford and succeeded in the industry. Angel also has a clear connection to the life of Tracey Africa Norman, a transgender model who rose to prominence in the fashion industry and was cast out. Carmen Xtravaganza was a trans model who achieved modeling success in Europe, just as Angel does at the end of season 2. Willi Ninja was a fashion dancer, house father, choreographer and model who lent his story to Damon. Elektra Abundance is inspired by Crystal Lebeija, down to the cadence of her voice. Dorian Corey was a drag queen and may also have figured in Elektra's story; a mummified body with a gunshot wound to the head was found in her apartment, believed to be an abusive client or boyfriend. Candy Ferocity was probably inspired by Venus Xtravaganza, a transgender sex worker who was harassed and later found murdered in a New York hotel.
